PHENOTYPIC AND GENETIC DIVERSITY OF NATIVE RHIZOBIUM ISOLATED FROM ROOT NODULES OF LEGUMINOUS PLANTS GROWN IN RECLAIMED SOIL, EGYPT

An investigation of the phenotypic characteristics 19 rhizobial strains, isolated from root nodules different plant legumes grown in soil agriculture farms Ismailia governorate, transpired. Most isolates were creamy or white opaque, mucoid, with a convex elevation, translucent, and smooth margined. Microscopic revealed that all bacterial rod-shaped had no positive affinity for Gram-stain. Identifying cultures any contaminants employed confirmatory tests based on prepared special media, including YMA supplement Congo red, glucose peptone agar, Kit-lactose Hoffer’s alkaline test. Based an infectivity test, proved their ability to reinfect host. These isolates, classified into two categories, included fast slow-growing rhizobia according growth YEM medium containing bromothymol blue (BTB). Meanwhile, assessment genetic diversity among these proceeded using ISSR RAPD markers, which marker more powerful tool discriminating tested than marker. The cluster analysis, marker, main groups. first group (1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 13), while second contained (12, 14, 15, 16, 17, 18, 19). Moreover, markers also showed groups diverse categories; 1 12 19.
